AP2-associated protein kinase 1 (AAK1) is a serine/threonine protein kinase that regulates clathrin-mediated endocytosis. It phosphorylates the AP2M1 subunit of the adaptor protein complex 2, enhancing its binding to cargo membrane proteins during endocytosis. AAK1 localizes to clathrin-coated pits at the plasma membrane and presynaptic terminals in neurons. It interacts with clathrin, AP-2 complex, and NUMB, playing roles in membrane organization, Notch signaling, and protein localization. AAK1 is implicated in viral entry processes for hepatitis C, Ebola, and Dengue viruses, making it a potential target for antiviral therapies.
