Recombinant Human Alpha-synuclein is a neuronal protein that plays critical roles in synaptic activity, particularly in the regulation of synaptic vesicle trafficking and neurotransmitter release. As a monomer, it enhances vesicle priming, fusion, and dilation of exocytotic fusion pores by increasing local Ca2+ release from microdomains. In its multimeric membrane-bound state, it functions as a molecular chaperone, assisting in the folding of synaptic fusion components (SNAREs) at presynaptic plasma membranes in conjunction with cysteine string protein-alpha. Alpha-synuclein also regulates dopamine neurotransmission by modulating dopamine transporter activity. Genetic alterations in SNCA resulting in aberrant protein aggregation are associated with several neurodegenerative disorders, including Parkinson's disease and Lewy body dementia.
